The vector does not have convenient restriction sites close to the insert. A BamHI digest yields 3 fragments from the insert, 5' 0.7, 3.54, 0.52 kb 3'. The end fragments contain vector sequences, and the vector band is about 3 kb. The insert contains the following restriction sites (nucleotide position according to GenBank J02635): BglII--7; BamHI--576, 4116; PvuII--637, 2174, 2536, 3040; HindIII--1440; XbaI--2217. Restriction digests of the clone give the following sizes (kb): BamHI--3.5, 3.0, 0.6, 0.5; EcoRI--4.6, 2.6, 0.4; PstI--4.3, 2.0, 1.3; HindIII--5.6, 2.0; SalI--7.6. Contains the complete coding sequence (nt 64-4482). |
