ATCC® VR-1479™ (M-varient) is a cell culture passaged linage of the M-varient (ATCC® VR-1314™) reported to induce diabetes in selected strains of mice such as Balb/cBy and DBA/2 mice. Note that virus loses diabetagenic properties upon multiple passage in cell culture.

Growth Conditions
Temperature: 37°C Recommendations for Infection: For best results, infection should be performed on a 75-80% confluent, 18-48 hour old cellular monolayer. Prepare dilution of virus in minimum amount of volume (e.g. 1 mL per 25 cm2), optimized for propagation (e.g. MOI 0.01-0.1 recommended). Wash monolayer two times with PBS or serum free medium prior to inoculation. Adsorb virus dilution for 1 hour at 37°C in a humidified 5% CO2 atmosphere, rocking every 15-20 minutes to redistribute inoculum. End adsorption by adding virus growth medium. Incubation: 4-10 days

ATCC® VR-1479™ was created by serially passaging ATCC® VR-1314™ two times onto NCTC clone 929 (L-929) cells, ATCC® CCL-1™. The original strain was isolated from the heart of mice inoculated by peripheral route with EMC of swine.
